Jake Gyllenhaal to play Mysterio; Michael Keaton returning as The Vulture

Following the news that Jake Gyllenhaal is in talks to play the classic Spidey villain Mysterio, it now looks like Keaton is coming back for the sequel

Could this possibly mean the Sinister Six is coming? I would bet this sequel will indeed show the early formation of them. But that being said, finally seeing Mysterio onscreen is exciting news and Jake’s casting is excellent. Can’t wait to see the two share the screen.

The only logical thing here is to start the movie during the filming of Tony Stark's biopic starring Simon Williams. Quentin Beck does something to sabotage the movie, Pete's already hanging around set because it's about his pal Tony so Spidey's right there to come in and save the day, but Beck gets away, leading to his transformation into Mysterio.

I read that and thought similarly. The Vulture, Mysterio, and likely the Scorpion make for a nice half to a Sinister Six. Assuming they forego villains already featured in previous Sony Spider-Man films-- the Green Goblin, Dr. Octopus, the Sandman, Venom, the Lizard, the Rhino, and Electro-- who do you reckon will make up the other half of the Six?

The Prowler was hinted at in the first film, but makes for an unlikely addition, unless he's drafted in by Toomes, perhaps only to later turn to Spider-Man's side? The Shocker could be in the mix as well, leaving only one spot unfilled. Kraven is long overdue for a cinematic appearance, but he likely warrants top villain billing in a future film, so my money is on a "lesser" villain-- Hydro-Man, Cyclone, the Chameleon, or another C-Lister.

Last year it was reported that Sony had plans to develop spin-off films with Mysterio and Kraven. Now that Mysterio’s involvement in the Spider-Man sequel has been announced, I believe Kraven being in this upcoming Spider-Man film isn’t unlikely. That is of course assuming we’ll be seeing the start of the Sinister Six, so at the very least he might be hinted at.

As for those who might potentially make up the rest, it’s hard for me to say. Unless they decide to add iconic villains we’ve seen on screen already (Doctor Octopus, Green Goblin, Sandman, etc), literally any Spider-Man villain could be appearing in this. Shocker is a likely candidate.
