It doesn't make sense since he seemed so hell-bent on finding and murdering Jews. If he was that obsessed with murdering every single Jew across the French landscape why let her go (and laugh about it)?

COL LANDA Oh, I don't think the state is in too much danger, do you?

COL LANDA I'm glad you see it my way. Besides, not putting a bullet in the back of a fifteen year old girl, and allowing her to escape, are not necessarily the same thing. She's a young girl, no food, no shelter, no shoes, who's just witnessed the massacre of her entire family. She may not survive the night. And after word spreads about what happened today, it's highly unlikely she will find any willing farmers to extend her aid. If I had to guess her fate, I'd say she'll probably be turned in by some neighbour. Or, she'll be spotted by some German soldier. Or, we'll find her body in the woods, dead from starvation or exposure. Or, perhaps-she'll survive. She will elude capture. She will escape to America. She will move to New York city. Where she will be elected, President of the United States. The S.S. Colonel chuckles at his little funny.
While what you say comes from the script, it doesn't necessarily negate what Calvin says above, that Landa let her get away for sport, enjoying his power over life and death. She presented no danger to the state and would either die in the woods, be turned-in by frightened French people or escape to another country. Whatever the case, it was fun for Landa to decide on a whim to let her go, not to mention it would keep his business alive and interesting.

Well, Col. Landa was an unbelievably arrogant individual who made it very clear the amount of disdain and contempt he had for the people around him in almost every word he says to another person. It isn't until the final moment that he's outsmarted by someone "intellectually inferior" who managed to hit him where it hurt most. Of course he'd never believe a teenage French Jewish girl could manage to come into a position of power that would allow her the ability to punish the entire Nazi high command.
As for Landa killing her personally. She was running full-tilt and by the time he could get off an aimed shot she was already 40-50 yards away, every second he hesitated bought here another 5 yards. Hitting her at 40-50 yards would take an excellent/lucky pistol-shot, by the time he looked like he might shoot she was probably closer to 100 yards away.
It would have diminished his reputation if he shot several times and missed her completely, which was the very likely outcome. Of course, they could have pursued her and likely gotten or shot her, but at that point, he probably did not think it was worth the trouble and she'd probably turn up anyhow...
