I think a lot of the best quotes came from the Tick's little moral at the end, the best of those being:

To this day that line can bring a smile to my face. Pure fried gold.

cuts in dream sequence to the interior of the UN as Speak, dressed in a cape, is addressing them in French.
Tick (offscreen) - "Speak. Come here, boy!!"
Speak - "I'm sorry. I have to go. The Tick needs me."
Speak flies off into the sky and lands next to the Tick standing on a cliff watching Arthur being attacked by a six armed monster or something like that.
Speak - "Well, it looks like Arthur's gotten himself into a bit of trouble again."
Tick - "Sure has, Speak."
Speak turns to the Tick - "I Love You."
I almost pissed myself the first time I saw that.
- "Is Wayne Brady gonna have to choke a bitch?"

In the event of a city emergency, you have but to aim this beam at a conveniently passing cluster of clouds and we will speed to the scene dispensing justice left and right.
Mayor Blank: You know, we have your phone number.
The Tick: Also useful.
Mayor Blank: The thing is, boys, we have a lot of these gizmos and they just don't work. Every time we flash the Die Fliedermaus signal, he disconnects his phone and leaves town for a week.
They did it in the first episode of the cartoon. The Tick even makes a menacing sucking sound (well, if there is such a thing) through the straw at the man.
