A video surfaced of a shoot in Italy which featured Esai Morales' villain and Hayley Atwell's character fighting each other, so there could be a connection between the two characters, McQuarrie has hinted that Atwell's character would be in the middle meaning she isn't on the good or bad side.

As for the other new characters, Morales is obviously playing the villain (a role that was originally going to be Nicholas Hoult's), Atwell, could be playing an ally of sort's to Ethan based on the fight video, Pom Klementieff and Shea Whigham, dunno what characters they're playing but McQuarrie hinted that Klementieff would play a villain last year, but has been seen filming with Rebecca Ferguson, Cruise and Simon Pegg in the last few weeks, same with Whigham.
