Can someone explain the significance?

It's a reference to cons, fake psychics, who start believing they have legitimate powers.

Because to get the full effect, it's best to watch the show with one eye shut. Also the show was originally called Shut Eye For The Straight Guy, but they dropped the last part because they thought it might confuse some people.
