I think it's established that Alex Wesker will have to be a primary antagonist in the next major game.
The big question is, why did he betray Spencer? I read the Wikia page, and I think it has something to do with George Trevor, the guy who helped found Umbrella and was supposedly killed by Spencer.
I'm sure he was the unidentified executive from Umbrella Chronicles, and I suspect he will be the 'secret' villain in the final RE game.
Seeing as George Trevor is the bearded crimson head in the mask room in the Gamecube RE, I'd say that is highly unlikely.

To add on to this, he's the bearded Crimson Head whos mouth Chris stuffed a grenade into, then put a bullet in it, which promptly removed the head completely which, according to zombie law, means it's very very VERY permanently dead. That and the mansion blew up with his corpse still inside, so if removing the head didn't do it, that sure did.
