Was that mind reading trick a plant? If not, how did they know what letters she was going to pick?

She wasn't a plant. The trick was done with what is called "pre-show". Pre-show is something the magician does before the act. There are plenty of ways to get the information. Something called "dual-reality" might have bee used as well.

Before the act, the mindreader asks for volunteers. It might be done openly in front of the entire audience or they might approach one person. Without going into a bunch of detail and different ways it could be done, the mindreader does a trick or series of tricks with someone and gets information from the volunteer. It's done under the guise of other tricks so that the volunteer doesn't know exactly how the mindreader got the info but if you spoke to the volunteer you'd find out it wasn't the first time they spoke to the mindreader.

No, there's no need for a plant or even a "pre-show". The trick they did is a variation of an old trick called a book test (really none of their tricks were anything original, but they did a good job of combining them all into a coherent act).

I believe the OP is talking about the trick were the woman was told the initials of the first person she kissed (TZ).

A book test is where a spectator chooses a page of a book, looks at a word (usually the first on the page) and the magician can tell what word the spectator looked at. I'm not sure what you're saying that has to do with the trick the OP referred to.
