I believe the grammar for which you are searching is "Gru is based on whom?"
Folks forget "Whom" is a wonderful objective case that is woefully under used.
And WHOM are you to tell me how I should write it??
Ok, I'm kidding, sorry, English is not my first language, and I often translate it from my original language. But, wouldn't it be more correct like "who is Gru based on?". I mean, like I had it, but without "from"?
Dream, if you had said "Gru is based on whom?" you would have been technically correct . . . and everyone would know you are not a native English speaker because noboby but nobody ever uses "whom," ever.
"Who is Gru based on?", while technically incorrect, because you should never end a sentence with a preposition, is the way everyone would say it.

Whom exists, but it is very stiff and formal. Casual language does not use whom. People who do use whom are made fun of.
The best way to ask the Gru question is, "Who is Gru based on?" This is true, even though that sentence ends in a preposition. No one would correct you for that "mistake".
