why John Nolan (?) before he shoots Hunter, says " five men, five guns; now do you get it?"

I haven't seen it in a while but is he referring to the death squad that went to the Karachi Club and indicating that he was a member?

Yes, he was one of the five who went into the Karachi Club to "clean up" after Eddie whacked Dawson.

Near the beginning of the film Hunter and Nolan are discussing the cartridges which came from five guns, although witnesses alleged there were only four shooters. They both said words to the effect of "I just don't get it" with Nolan obviously pretending to be baffled at that stage.
