How did alice know? -spoiler-

When saya is almost dead, alice gives her her own blood.. BUT, how the flying f. did she know saya was a vampire, she hasn't said anything to alice about it only AFTER alice had given her blood.

Because Alice is not a moron and used her brain. It was especially easy for her to figure it out because of all the vampires seen. Had Alice remained in the dark about the vampires, she never would've found out Saya was one.

Because earlier Saya told her the bloodsuckers "provide her with what she needs" and probably saw the bottle of blood in her room before they were attacked by the sniper.
