Well, if we have to keep it to a single line, I think the best was said after Diego and Sid had just been completely humiliated by Crash and Eddie.
Diego: "If anyone asks, there were fifty of 'em... And, uh... they were rattlesnakes."
But the best exchange was between Manny and Ellie.
Ellie: "Oh, now I understand why the possum boys didn't find me appealing."
Manny: "That's too bad, because as far as mammoths go, you're, uh... you know."
Ellie: "What?"
Manny: "Um... uh... well... attractive."
Ellie: "Really?"
Manny: "Sure!"
Ellie: "What about me is attractive?"
Manny: "Huh? Well, oh, well, I don't know, um... uh, well... there's your... uh... butt?"

I can't remember any specific lines, but the scene where Sid enters the tribe is great. Me and my family laughed our heads off! Whenever I feel sad I think of Sid, his funny eyes and that scene - then I'm all laughs again!

The Hands Down, BEST line in the entire film was Manny's confession to Ellie;
"Ellie, I don't want us to be together because we have to, I want us to be together because we want to, and I want to be with you Ellie... So what do you say?"
I know it's not really funny, but it still is in my opinion the best line of the entire film... but here are my top five fav funny lines;
1. Little Start Girl - "Sometimes I Throw Up!"
2. Sid - "Stop Hey, What's that Sound? All the mammoths are in the ground!" "If your species won't continue clap your hands!"
3. Ellie - "He's fat."
4. Manny - "Fine, the wild ass boy went home to his wild ass mother!"
5. Crash - "I believe I can fly!"
