Where does that recurring hopalong harmonica riff come from? It sounds so familiar, but I just can't place it.

it got me too. i had to pause the movie to hum it to try and place it. this might be really off the mark, but i think it's a theme song from a television show that i watched as a kid. the odd couple, played western style?

I think it was a commonly used background tune used in the Trinity movies.
“They call me Trinity” “Trinity is still my name” etc.

Go to any of the spaghetti westerns from Clint Eastwood to Sergio Leone and there is a similar musical theme through them.
In Once Upon a Time in the West, the harmonica music is thematic with Charles Bronson's story. It lopes along throughout the movie. However, I don't want to say any more. That would create a spoiler.

It sure as hell sounds like a morricone-ised version of "The Odd Couple" theme to me! Maybe they couldn't get the rights to use some of the classic spaghetti-western themes?
