shareNo, The Rock did not screw up Doom. The Rock had no say in the plot or story, or the visuals or anything but his character. IF ANYTHING about Doom was GOOD, it was The Rock as Sarge, and Karl Urban as Reaper. Everything else was atrocious.
The Rock, John Cena, Brock Lesnar... They all could do an interesting version of Duke.
Personally, I think Kurt Russel circa 1999 would have been fantastic. Just after he made Soldier he was buffed up and his hair was perfect, not to mention his attitude and acting skills often allowed him to change his personality and voice. He would have been an awesome Duke.

It should be done, what about Ron Perlman to play the title role. He was good as Hellboy. Who else? And what about those strippers, I mean, you could have like loads of cameos since its a comedy. Just to please the critics, what if there was a sub-plot where he actually takes it upon himself to father and foster an underage stripper, played by Scarlett Johansen possibly (well it would make a good comedy). The monsters too, they need to be butt ugly, shotgun/chaingun wielding maniacs. Personally, not this specific idea, but a film would work.

If they do it they better do it right, I'm sick of Video Game films ignoring the plot of the games their based on. Start from this one work up to 3d.
The tone of the Movie should be across between Army of Darkness and Commando. Over the top satire of 80s 90s action, horror sci/fi flicks.

Its freaking terrible for film. You would have to change so many elements to make it work, not only with what most people associate with Duke, but you'd also be changing to the point as to WHY BOTHER? Dr. Proton as a villian is so nails-on-a-chalkboard stupid.
Whenever someone thinks of Duke. They think of Babes, Attitude, Explosions, Pig Cops, and Alien Scum. and thats exactly what Hollywood will go with.
Its gonna be similar to Hellboy or Blade when the time comes. A semi reimagined backstory and an entirely original plot with a lot of the same elements. (Aliens, Babes, Attitude, Violence.)
Yea thats what I would do to. Base it on The first platform game where Duke fights robots and his nemesis Dr. Proton in a ravaged city.
I'd mix in ideas from Manhattan Project too as in many ways it is a kind of remake.
That would give us Cyborgs and Mutants for Duke to fight but might disappointed those who think Duke Nukem 3D is the first game.
