In a word no.

Asenath was the daughter of the priest Potiphera (not Potiphar) from Heliopolis (On) and was given to him (as his wife) as a gift from Pharoah from interpreting his dreams about the cows and the ears of corn. (Genesis 41:45) This passage is the first time she is mentioned in the bible. The other occurances happen when she gives birth to Manasseh and Ephraim (depending on which version of the bible you read: NIV or King James, etc) and is mentioned as part of Jacob's tribe/family (Genesis 46:19).

The events in Potiphar's house are in Genesis 39 and is in the prison during Genesis 40. This is well before when Asenath is given to Joseph as his wife.

No. She was the daughter of Potiphar. Making her his niece is yet another example of the ridiculous and unnecessary liberties that were taken by DreamWorks.
