Is this the episode that had a character parodying Katharine Hepburn? If not, which episode is it?

I know this is late to the game, but their version of Elizabeth I was a parody of Hepburn.

You got it! Or, more to the point, she was a parody of Hepburn as Elizabeth I, since Hepburn played her in movies.

Hepburn never played Eluzabeth 1 the only Royal Tudor she ever did play was Queen Mary Stuart in The movie Mary of Scotland who was Elizabeth 1's cousin. Bette Davis played Elizabeth 1 in at least 2 movies I can think of.
