I think it could be done as maybe a 2-part TV-miniseries type of thing which would revisit the characters 25 years later, and, if the reception was good, possibly set up a "Next Generation" type of thing with Holling/Shelly's daughter Miranda as the new main character.
Maybe have the storyline have Randi as a brilliant med student who went off to med school in California and was all set up with a residency in LA, but then happened to meet Joel Fleischman, now a successful practitioner with 20 years of practice in Manhattan behind him, at a conference in Chicago.
One way or another the wheels are put in motion and Dr. Randi returns to Cicely to take over operation of the Alaskan Riviera Medical Clinic.
It wouldn't depend on any of the original cast making long-term committments to the show -- they could be written in (or out) depending how much time any individual actor wanted to commit.

I think the 'where are they 25 years later' thing is very interesting. There would be MANY changes to the town, though, I don't know if I'm prepared to see it lose it's rustic-ness. Cicely was a dream world. I know in the real world, it would not have been some whimsical and charming, full of kind, odd, silly characters.

When I first saw the Film Farms Crowdfunding site several days ago, it was in it's 2nd last day of 7 day campaign. And it seemed to have received only about 1/4 of the goal. But yesterday I saw the site again, and it now has 3 weeks more to go in the donations campaign, and was up to almost $45,000 which is approaching half of the goal. So the donation effort seems to be accelerating.
Meanwhile, I don't get regular or frequent notices of messages posted about northern exposure, here on imdb. And after being gone from this message board for some months, it was hard to find it again. Any tips on imdb message board search for northern exposure, would be appreciated.
