They should never do a remake

But if for some reason Hollywood feels compelled to do so I can see Brie Larson and Alexander Skarsgard playing the leads.
Oscar Isaac would do an amazing douchebag interpretation of Humperdinck.
Something tells me Chris Pratt would make a pretty okay Inigo.

But please, film industry, don't touch this one.

Agreed

I literally only got into The Princess Bride a few days ago and it's been less than an hour since I saw the film for the first time so I have no nostalgic connection to it, however I wouldn't be opposed to a remake.

I don't think it would be the same but there were some scenes from the book that never made it to film including more detailed backstories of Fezzick and Inigo (who are my favourite characters, in fact I would totally read/watch a spin off about them). It could work as long as they didn't try to impersonate this film. If they took their own approach it could have the potential to be good.

No. The movie is perfect just the way it is. It should never be remade or updated or changed in any way.
