Did Nicholson miss the wets in the back of the truck/van?

You are incorrect. If you watch the scene again you will see that while Charlie was going after the driver Manuel came to the van with the two "wets". In other words, Manuel locked them in the back of the van after Charlie had checked the van and then took off for the driver.

Cat (Harvey Keitel) set it all up. In the scene just before the one in question, Cat said to Manuel, " I'm gonna stick ol' Charlie so deep in my pocket they're gonna have to pipe light into him".

They were planted after. It was a plan setup by Keitel's character to prevent Nicholson from spilling the beans on the operation. After he chases the driver away, another car pulls up and two mexicans get in the backseat of the truck. The person who is putting them in the truck says "Una hora", leading them to believe someone will pick them up in the hour, only to leave them in the blistering sun all day. Nicholson was set up.

A day without a buzz is a day that never was.
