I'm not sure if this qualifies as a plot hole, but it is something that has always bothered me: At the end, all the townspeople gather round to feast on the roasted cadaver of Queen Carlotta. However, earlier when the gang burst into the Queen's chambers, the rabid Princess Coo-Coo bit the Queen on the leg, thus transferring her disease to her mother. So unless the rabies virus is killed by high heat, such as that used in roasting, all the townspeople who ate part of the Queen are now infected with rabies, which was the Queen's plan all along. Ironic.
jasonhurd, I was just thinking about it even though I just finish Hairspray. I do think it's ironic. The Carlotta-Ala-King looks appetizing. I wonder if it's stuffed.

Liz Renay mentioned it in the commentary, then I stopped and thought about it, thinking "PLOT HOLE!" Then, when a poster mentioned its irony, it all became clear. Waters was into irony, and I think he did this purposely.
