Guest who went on to have the biggest career.

Robert Redford from Nothing in the Dark - 1962
Robert Duvall from Miniature - 1963

Burt Reynolds - The Bard
Ron Howard - Walking Distance (I'm also considering his directing career.)

Shatner

And who had the biggest career prior to appearing on The Twilight Zone?

Buster Keaton?

reply

I'd have to say William Shatner due to his longevity and his ability to stay relevant up into his 80's. Robert Redford, Robert Duvall, Burt Reynolds all had great moments but after a decade or two of being hot movie stars they mostly fizzled out.

That's true. While initially one would probably say Redford or Reynolds as they became above the title A list movie stars of the 70s (and the 80s, and occasionally into the 90s, 00s and even today with Redford in Winter Soldier etc) and had some truly classic movies like Butch Cassidy, Deliverance etc , Shatner with Captain Kirk/Star Trek is just absolutely iconic.. I mean even today Kirk/Shatner is still there and still popular .. even the new Trek movies with their huge budgets/recast couldn't really get out of the original Trek and Shatners shadow as its just too big

Charles Bronson is a contender

I have no choice but to say Billy Mumy. And it's good that he had the biggest career. Real good.
