Since they talk about Poughkeepsie, and actually go there to the rest home, they have to be living in NYC.

They seem to have moved there between Shadow of the Thin Man and The Thin Man Goes Home. In the former, they're still living in San Francisco (but in an apartment rather than that great house they had in After the Thin Man), and while we never see them at home in the latter, home seems to be New York -- Sycamore Springs is on the East Coast somewhere and they take a short day train trip to get there, Nora says Nick's parents have never come to visit them in NY, etc.
Wasn't that the Empire State Building in the view from their apartment? Also, the sign on the harbor boat said New York Police.
