...Essex smacks Elizabeth in the rear and they both laugh together on the steps. I crack up with them every time I see that part.

Flynn and Davis seem to be genuinely laughing. I've read they didn't get along very well, but I like to think for this one moment they both found something hilarious they could use for the film.

Who knows whether it's true, but that thought makes that scene more enjoyable to me.

In "My wicked, Wicked Ways" Flynn explained it as the stage direction was that he embarrassed the Queen.

He could only think of smacking her bum as a way of embarrassing her. He claimed that Davis told him he would have to do it again until he got it right! Flynn finished up feeling mortified by it.
