I think it was more emotionally based affair...she was neglected by her workaholic husband and she found friendship with the man next door. She did take it too far by basically deciding on a divorce and not even considering her little girl's feelings.

(I just watched this for the first time on Amazon Instant Video...rented it because it was one of the Shirley Temple (RIP Curly Top!) movies that I hadn't watched before.)

Did the nurse hope that the husband was going to leave his wife and she would marry the doctor?

I don't know about you, but I thought it was the weakest Shirley Temple movie I've seen so far. There's still a few I haven't seen. Like a few of the later ones when she was a teenager.

I found it a bit rushed the mother was going to marry the other guy, she had only known him for five minutes.

That happens on all the movies of the 1930's it seems. A man and woman don't' even need to have a date or a first kiss. I've seen movies where 2 people just share one hug and that's enough to run off and get married.
