Wasn't TOS where the Prime Directive came up first? I seem to remember Kirk being the one to invoke it on several occasions.

Yes the Prime Directive is mentioned in TOS. I'm not sure of all the episodes that mention it but "Bread and Circuses" mentions it. Saw it this weekend during BBCAmerica's Trek-A-thon The character Claudius taunts Kirk with it a couple of times. "Oh there's that darn Prime Directive again."
